Zenit AMIDOT

The ZENIT•AMiDot test is based on the Immunoassay principle. It consists of a Reagent ZENIT•AMiDot Reagent Kit containing conjugate, sample diluent and wash buffer, and a ZENIT•AMiDot Slide Pack containing glass microscope slides with 4 or 8 wells defined by a hydrophobic mask. These wells are coated with an array of autoimmune antigens, calibrators and controls.

Each ZENIT•AMiDot panel-specific slide can assay 4 or 8 patients, one patient per well

Each well contains:

  • a 9 x 9 array of up to 38 different antibodies or antigens measured in duplicate
  • calibration curve for each antigen
  • assay controls

ZENIT•AMiDot slides ensure quantitative, reliable and reproducible results

  • An anti-human IgA status control (a negative result may indicate IgA deficiency)
  • An anti-human IgG status procedural control
  • Intra well calibration for IgG and IgA provides patient-specific calibration
ZENIT•AMiDot slides can be run either manually or on automated IFA slide processors

The principal steps of the method are:

  1. Serum sample dilution (1/80) and incubation with autoimmune antigen array fixed on glass slides for 30 min in a moisture chamber at RT
  2. Incubation with flurophore conjugated goat anti-human IgG/IgA for 30 min at RT
  3. Assessment: automated analysis of slides

Signal intensity is directly proportional to the amount of antibody present in the sample. Results are expressed in U/ml or IU/ml.

ZENIT•AMiDot slides may be read as soon as they are prepared. However, no significant loss of intensity occurs if reading is delayed for up to 48 hours. In this case, ZENIT•AMiDot slides should be stored in the dark, at room temperature.

ZENIT•AMiDot Fluorometer Automatically scans the slide for each patient sample

  • Automatic scan of the bar code of the slide
  • Analyses each array dot using an algorithm that uses the built in quality controls and calibrators in each well
  • Produces a report for each patient/well
  • Takes less than 40 seconds per 8 well slide

Quality Control of ZENIT•AMiDot system

The internal controls built into the ZENIT•AMiDot array ensure that each patient sample has been diluted correctly and that the test procedure has been followed.

ZENIT•AMiDot Fluorometer will interpret the microdot array and produce a report listing concentration and positive, borderline or negative status for each antigen if the calibration and controls meet required specifications.

If the internal controls do not meet specifications a report listing the control failure is generated and no patient results are displayed.
